研究内容
オートファジーは多くの疾患と関わりがあります。私達は、オートファジー・リソソーム分解系と神経変性疾患の関わりを研究しています。
図1 様々な疾患にオートファジーは関与している
脳神経細胞において細胞の老廃物を分解するオートファジー機能の低下はパーキンソン病、アルツハイマー病、バッテン病などの神経変性疾患と深い関わりがあります。
特にオートファジー関連因子のなかで唯一の膜タンパク質であるAtg9と、リソソームの主要タンパク質分解酵素であるカテプシンD, B, Lに注目して、脳神経におけるオートファジー・リソソーム分解系の研究をおこなっています。Atg9を脳特異的にノックアウトしたモデルマウスでは、神経変性疾患様表現型を示します。
カテプシンD脳特異的ノックアウトマウスでも神経変性疾患様表現型を示し、オートファゴソーム形成(分解する細胞質成分をオートファゴソームに包み込む現象)から、リソソームによる分解(オートファゴソームに包み込んだ細胞質成分をリソソームの加水分解酵素により分解)まで、いずれの段階でも神経細胞維持には必須であることがわかります。また、カテプシンDの遺伝子は、神経セロイドリポフスチン症の原因遺伝子の一つ、CLN10、でした。

現在、これらのモデルマウスを使って、加齢・神経変性疾患におけるオートファジー・リソソーム分解系の機能について、解析を行っています。
また、細胞・組織の形態的な解析の充実を図るため、新規のCLEM法(光線-電子線相関法)の技術開発研究も行っています。
